This advanced course examines the molecular basis of cancer and the principles underlying anti-cancer therapy. It covers the hallmarks of cancer, oncogenes and tumour-suppressor pathways, genomic instability, the tumour microenvironment, invasion and metastasis, and cancer stem-cell biology. It then reviews the rational basis and mechanisms of action of conventional, targeted, immunological and emerging precision therapies, together with the molecular determinants of therapeutic resistance and the translational pipeline from target discovery to clinical application. Emphasis is placed on interpreting cancer genomic data and appraising preclinical and clinical evidence to inform precision-oncology strategies
